摘要
目的:探讨高血糖对抗结核药利福平、异烟肼的药动学影响,为糖尿病合并结核病治疗提供理论依据。方法:用高效液相色谱法分别测定糖尿病合并肺结核患者血糖控制前(高血糖时)、控制后及单纯肺结核患者抗结核药利福平、异烟肼的血清药物浓度,采用大型药理学计算软件(DAS2.0)处理血药浓度数据并计算参数。统计学分析血糖控制前、后抗结核药物的药动学,并分别与单纯肺结核患者进行对比。结果:经比较,各药动学参数(AUC0→t、AUC0→∞、t1/2、Cmax、tmax)在不同类型患者间的统计学差异无显著性(P>0.05)。结论:糖尿病的并发以及糖尿病水平的控制对于利福平、异烟肼在人体内的代谢没有显著性影响。
OBJECTIVE To study the effect of high serum glucose on the pharmacokinetics of two anti-tuberculosis drugs,isoniazid (INH) and rifampicin (RFP). METHODS The levels of serum INH and RFP were measured by high performance liquid chromatography (HPLC) hefore and after the control of serum glucose in pulmonary tuberculosis patients with diabetes. The data were calculated by soft of pharmacology DAS 2. 0. The pharmacokinetics of INH and RFP were analysed before and after the control of serum glucose, and compared with patients without diabetes, respectively. RESULTS There was no significant difference in the pharmacokinetics parameters such as AUC0→∞、AUC0→∞、t1/2、Cmax、tmax(P 〉 0. 05) between the two groups. CONCLUSION The pharmaeokinetics of INH and RFP were not affected by the factors of diabetes and levels of serum glucose.
