干细胞源胰岛素分泌细胞中胰岛素原转化障碍的原因及对策 被引量:1

The obstacles in the conversion of proinsulin to insulin in stem cell-derived insulin-producing cells

摘要 近年来,诱导干细胞向功能性胰岛素分泌细胞分化已取得了较大进展,但不论何种干细胞来源,采取何种诱导方案,诱导的胰岛素分泌细胞均不同程度地存在着胰岛素原向胰岛素转化的障碍,严重影响了其治疗糖尿病的效果。该文从胰岛素合成和分泌的生理学角度探讨胰岛素分泌细胞不成熟的原因及机制,从而指出目前在干细胞向胰岛素分泌细胞诱导过程中存在的不足并提出相应的解决办法。 Progress has been made in inducing stem cells to differentiate towards functional insulin-producing cells (IPCs) in recent years. Despite the sources of the stem cells and the protocols used for induction, there are still difficulties in the conversion of proinsulin to insulin in the stem cell-derived IPCs, which hamper the IPCs' effects on diabetes therapy, This review discusses the possible causes and the underlying mechanisms from the physiological viewpoints of insulin synthesis and secretion, and points out the limitations and solutions in the course of inducing stem ceils towards IPCs.
