摘要 目的 研究β-淀粉样蛋白(Aβ)对大鼠海马内Fyn和NADPH氧化酶表达的影响及其相互关系,探讨Fyn在阿尔茨海默病(AD)发病中的可能作用。方法筛选雄性SD大鼠45只,随机分为三组:模型组、PP2处理组、对照组,通过立体定位技术将Aβ1-42注射模型组大鼠的双侧海马;PP2处理组先注射PP2,1h后再注射Aβ1-42,对照组注射等容积的生理盐水。通过RT—PCR方法观察Aβ1-42对大鼠海马区内FynmRNA和NADPH氧化酶mRNA表达的影响,并分析两者的关系。结果模型组大鼠海马内FynmRNA(2.08±0.45)和NADPH氧化酶mR—NA(1.40±0.19)表达较PP2处理组(1.40±0.24和1.06±0.13)及对照组(分别为1.10±0.15和1.02±0.11)均有明显升高;且海马内FynmRNA和NADPH氧化酶mRNA水平表达具有显著相关性(r=0.467,P〈0.05)。结论Aβ诱导大鼠海马内Fyn和NADPH氧化酶表达增强,同时两者表达呈正相关关系。提示两者均参与了Aβ诱导的炎症反应过程,Fyn可能是NADPH氧化酶表达的介导因子之一。 Objective To investigate the expression of Fyn and NADPH oxidase in the hippocampus of rats and their correlation, and to the contribution of Fyn to Alzheimer disease pathogenesis. Methods The animal model of AD was established by Aβ1-42 stereotactic injection into bilateral hippocampus of rats. The expressions of Fyn mRNA and NADPH oxidase mRNA in the hippocampus were detected by RT - PCR ( reverse trascription - polymerase reaction) ,and the relationship of Fyn mRNA and NADPH oxidase mRNA was analysed. Results Injection of 2.5 μl of 500 μmol/L Aβ1-42 into bilateral hippocampus of rats induced:①AD - modelized rats showed the increased expressions ofFynmRNA[(2.08±0.45) vs(1.10±0.15)]and NADPH oxidase mRNA[(1.40±0.19) vs (1.02±0.11 ) ] ( all P 〈 0.05 ) as compared with control group. ②The positive correlation was found between Fyn mRNA value of gray scale and NADPH oxidase value of gray scale ( P 〈 0. 05, r =0. 467 ). Conclusions Aβ could obviously induce the increased expressions of Fyn and NADPH oxidase in the hippocampus of rats, and expressions are positively correlated. It suggests that Fyn and NADPH oxidase participate in inflammatory reaction induced by Aβ and Fyn probably is one of mediate factors inducing NADPH oxidase expression.
