血小板源性生长因子D基因多态性与尘肺易感性关系的研究 被引量:3

Study on genetic polymorphism of platelet derived growth factor D associated with pneumoconiosis sensibility

摘要 目的研究血小板源性生长因子D(PDGF-D,-858、3166)位点基因多态性与尘肺发病的关系。方法选择确诊的96例Ⅰ期尘肺病患者为病例组,以接触同性质粉尘,工龄相近的非尘肺的接尘工人90例为对照组。采集周围静脉血,应用聚合酶链-限制性片段长度多态性(PCR-RFLP)技术检测PDGF-D(-858、3166)基因多态性。结果PDGF-D(-858)基因型(AA、AC、CC)在尘肺组的分布频率分别为63.54%、17.71%、18.75%,对照组分别为57.78%、16.67%、25.56%。PDGF-D(3166)基因型(CC、CG、GG)在尘肺组分布频率分别为54.17%、23.96%、21.88%,对照组分别为63.33%、12.22%、24.44%。分析发现PDGF-D(-858)基因型分布频率在尘肺组和对照组差异无显著性(P>0.05),PDGF-D(3166)基因型在尘肺组和对照组差异有显著性(P<0.05)。结论PDGF-D(3166)基因多态性可能与尘肺发病有关联。 Objective To explore the relationship between platelet-derived growth factor D(PDGF-D,-858.3166) genetic polymorphisms and the pneumoconiosis sensibility. Methods Case group of 96 male stage I pneumoconiosis patients diagnosed by the pneumoconiosis diagnosis expert panel according to the Chinese national diagnosis criteria of pneumoconiosis( GBZ 70- 2002) were selected as case group. 90 workers exposed to same dusts without pneumoconiosis were selected as control group. The samples of peripheral venous blood were drawn from the subjects. The PDGF-D (-858 and 3166) genetic polymorphisms were detected by polymerase chain reaction restriction fragment length polymorphisms (PCR-RFLP) techniques. Results The frequencies of PDGF-D (-858) (AA,AC and CC)genotypos in case group were 63.54%, 17. 71%, 18. 75%, and 57.78%, 16. 67%, 25.56% respectively in control group. PDGF-D(3166) ( CC, CG and GG) genotypes in case group were 54. 17% ,23.96%, 21.88% ; 63.33% ,12. 22% ,24. 44% were respectively in control group. The statistical analysis indicated that there were significant differences in the frequencies of PDGF-D(3166)( CC,CG and GG)genotypes between case group and control group( P 〈 0.05). Conclusion PDGF-D (3166) genetic polymorphisms could be associated with the development of pneumoconiosis.
