层粘连蛋白及受体与肿瘤的关系

摘要 目前,恶性肿瘤是威胁人类健康最严重的疾病之一,肺癌是重要的死亡原因之一。在美国,每年约有17.5万肺癌患者死亡,肺癌的预后差,五年生存率仅17%。癌细胞首先通过整合素(层粘连蛋白的高亲和力的受体)与基底膜粘附,随后分泌多种蛋白溶解酶(包括Ⅳ型胶原酶、组织蛋白酶D、和尿激酶型胞浆素原活化物等)溶解细胞外基质成分,如层粘连蛋白(LN)、Ⅳ型胶原、纤维连接蛋白、蛋白多糖等,使基底膜产生局部的缺损,进而降解基底膜,穿透基质进入血液和淋巴管,从而导致血液及淋巴的转移,最后在一定的基质上定着、增殖,形成转移灶。由此可见,肿瘤细胞与各种基膜的作用对于转移过程具有决定性影响。
