IFN-γ对IL-2基因转染B16细胞治疗肿瘤的增强作用

摘要 本实验在完成IL-2基因转染瘤苗的基础上,探索了重组IFN-γ对其效果的增强作用。结果表明,IL-2基因转染使B16细胞成瘤性显著降低;小鼠经IL-2基因修饰的B16细胞免疫后,对接种的亲代B16细胞有保护作用;IL-2基因转染的B16细胞对接种亲代B16细胞的小鼠有一定的治疗作用,与重组IFN-γ联合应用后,治疗作用加强。上述结果表明,佐以合适的重组细胞因子,有可能提高单基因转染瘤苗的作用。 We observed the antitumor effects of B16 cells transfected with murine interleukin2 gene and the enhancing effect of recombinant interferonγ. The tumorigenicity of B16 cells transfected with IL2 gene decreased significantly as compared with parental tumor cells and mock vector transfected tumor cells. Moreover, inoculation of the IL2 gene transfected tumor cells activated cytotoxic T lymphocytes and induced protective immunity against parental B16 cells when combined with recombinant interferonγ, the therapeutic effects were enhanced. It suggested that IL2 gene transfected tumor vaccine combined with recombinant interferonγ might get better therapeutic effect, and it migth represent an approach for the immunotherapy for cancer.
