摘要
本实验在完成IL-2基因转染瘤苗的基础上,探索了重组IFN-γ对其效果的增强作用。结果表明,IL-2基因转染使B16细胞成瘤性显著降低;小鼠经IL-2基因修饰的B16细胞免疫后,对接种的亲代B16细胞有保护作用;IL-2基因转染的B16细胞对接种亲代B16细胞的小鼠有一定的治疗作用,与重组IFN-γ联合应用后,治疗作用加强。上述结果表明,佐以合适的重组细胞因子,有可能提高单基因转染瘤苗的作用。
We observed the antitumor effects of B16 cells transfected with murine interleukin2 gene and the enhancing effect of recombinant interferonγ. The tumorigenicity of B16 cells transfected with IL2 gene decreased significantly as compared with parental tumor cells and mock vector transfected tumor cells. Moreover, inoculation of the IL2 gene transfected tumor cells activated cytotoxic T lymphocytes and induced protective immunity against parental B16 cells when combined with recombinant interferonγ, the therapeutic effects were enhanced. It suggested that IL2 gene transfected tumor vaccine combined with recombinant interferonγ might get better therapeutic effect, and it migth represent an approach for the immunotherapy for cancer.
