摘要
本文用人HLA-DR3基因转染到小鼠黑色素瘤细胞系B16,构建了带有异种MHCⅡ类基因的B16-DR细胞。对此细胞进行免疫学分析发现,B16-DR细胞的致瘤性下降,免疫原性提高;经过B16-DR细胞免疫的小鼠可经受后续的B16细胞的攻击;过继免疫也可以在一定程度上保护小鼠抵抗B16细胞的攻击。上述结果提示。
B16DR cells with xenogeneic MHC class Ⅱ gene were constructed by transfecting the human HLADR3 cDNA into the murine melanoma cell line B16. The immunoassay for B16DR showed that the tumorigenicity of B16DR was reduced and the immunogenicity was enhanced, B16DR cells immunization protected the mice from wild type B16, adaptive transfection of the B16DR immunized splenocytes also protected the mice from B16 cells′ attack. These results suggest that MHC class Ⅱ gene transfection is favorable to enhancement of tumor immunogenicity.
