用FISH技术检测肝癌及喉癌细胞系染色体畸变 被引量:4

DETECTION OF Hep 2 AND HEPATOMA CELL LINE CHROMOSOMAL ABERRATION BY USING FLUORESCENCE IN SITU HYBRIDIZATION

摘要 目的探讨肝癌及喉癌细胞系染色体畸变。方法选用9种染色体特异性探针,分别对肝癌及喉癌细胞系的染色体进行荧光原位杂交检测。结果在两种细胞系中,这些探针均呈现异常的杂交信号。肝癌细胞系中主要是结构畸变,而喉癌细胞系中染色体数目畸变占主要地位。两种细胞系中染色体畸变所涉及的范围极广,而且畸变类型极为复杂。结论与常规的细胞遗传学分析方法比较,荧光原位杂交技术具有快速、准确、灵敏等优点。 Objective To study the chromosomal aberration of hepatoma cell line (1172) and laryngocarcinoma cell line (Hep 2).Methods Fluorescence in situ hybrization (FISH) was used with nine chromosomal special libraries.Results The abnormal signals indicated that very complicated aberrations existed in the two cell lines. This suggested that the aberrations also existed in other chromosomes not yet studies. In 1172, the major type was structure aberration, but in Hep 2, numerical abnormality was shown in almost all chromosomes studied.Conclusion Compared with traditional cytological methods, this technique has the advantages of being quicker and more accurate and sensitive.
