摘要
目的:探讨脂氧合酶抑制剂NDGA对人胰腺癌细胞株PCNA-1生长抑制、诱导凋亡及其对bcl-2、bax表达的影响。方法:采用MTT法检测NDGA对胰腺癌细胞株PCNA-1增殖活性的影响,流式细胞术检测NDGA处理的胰腺癌细胞株PCNA-1的细胞周期及凋亡相关蛋白bcl-2、bax的表达。结果:NDGA抑制胰腺癌细胞株PCNA-1的增殖活性,呈剂量依赖效应关系;细胞经100μmol/L的NDGA处理后G0/G1期细胞减少(P<0.05)、S期细胞增多(P<0.05),G2/M期细胞无明显变化;细胞经100μmol/L的NDGA处理48h后bcl-2蛋白与对照组相比表达下调(P<0.01)、bax蛋白与对照组相比表达上调(P<0.01)。结论:NDGA对胰腺癌细胞株PCNA-1有抑制增殖、诱导其凋亡作用,诱导细胞凋亡的机制可能与细胞凋亡相关基因bcl-2表达下调、bax表达上调有关。
Objective:To investigate the effects of lipoxygenase inhibitor nordihydroguaiaretic acid(NDGA) on the proliferation and apoptosis of PCNA- 1 and it's influence to bcl -2,bax protein expression. Methods:The effect of NDGA on the proliferation of human PCNA - 1 was detected by MTT assay, the effects of NDGA on the cell cycle of human PCNA - 1 and the express of apoptosis associated protein bcl - 2, bax were detected by flow cytometry. Results:NDGA inhibited the proliferation of human PCNA -1 as dose -dependent effect, deal with 100μmol/L NDGA the cell of G0/Gl stage decreased (P 〈 0.05 ), the cell of S stage increased (P 〈 0.05 ), the cell of G2/M stage was no marked change ;deal with 100μmol/L NDGA for 48h, the expression of bcl- 2 was down regulated compared with control group( P 〈 0.01 ), the expression of bax was up regulated compared with control group (P 〈 0.01 ). Conclusion:NDGA can inhibit the proliferation and induce apoptosis of PCNA -1 ,the mechanism of apoptosis is related to the down regulation of bcl - 2 and up regulation of bax.
