摘要
利用冷冻环玻璃化法对猪MⅡ期卵母细胞进行冷冻试验。结果表明:冷冻环玻璃化法冻后存活率达80.00%,显著高于OPS法和细管法的72.50%和53.33%;冷冻环玻璃化法激活后的卵裂率为7.22%,也显著高于后两者的4.85%和0;紫杉醇和细胞松弛素预处理能在一定程度上提高猪MⅡ期卵母细胞的抗冻能力,两种添加物对冻后卵母细胞的存活率和卵裂率无显著影响。
The effect of cryoloop vitrification on cryopreservation of porcine MⅡ oocytes was study. The results showed that the cryoloop vitrification method could make the survival rate of the oocytes reach 80.00% ,which was significantly higher than the OPS method(72.50%)and the straw method(53.33% ). The oocytes vitrified by the cryoloop vitrification method had a significantly higher cleavage rate (7. 22%)after parthenogenetic activation than those vitrified by the OPS method (4. 85%)and the straw method(0). The pretreatment of the oocytes with taxol or CB could partly improve the antifreeze capacity of the oocytes, and the two chemical additives had not a significant difference in the effect on the survival and cleavage rates of the vitrified oocytes.
