摘要
目的制备青黄珠口腔生物黏附贴片并考察其体外生物黏附性及释药性能。方法以青黛、黄连提取物、珍珠为药物,白芨胶、HPMC和卡波姆为黏附材料,通过生物黏附力和体外溶胀速率测定,优化青黄珠口腔黏附贴片处方。并对所制备的生物黏附贴片进行体外释放度和释药机制探讨。结果所筛选最优处方为白芨胶:Car-bopol:HPMC=1.5∶1∶0.5,药粉所占比例为25%;体外释放度试验证明不同阶段存在着不同的释药机制。
Objective:To prepare Qing-huang-zhu Buccal bioadhesive tablets and to observe the bioadhesive properties and drug release profile in vitro. Methods: The Qing-Huang-Zhu Buccal bioadhesive tablets were prepared by using Indigo Naturatis,extraction of Rhizoma coptidis and genuine pearl as model drugs, bletilla hyacinthine gum hydroxypropyl methyl cellulose (HPMC) and carbomer ( Carbopol 974P) as adhensive excipients , determining the adhesive force and swelling percent in vitro of which to optimize the formulation of Qing-Huang-Zhu Buccal bioadhesive tablets. Results: The Qing- Huang-Zhu Buccal bioadhesive tablets were prepared by bletilla hyacinthine gum,Carbopol 974P and HPMC with 1.5: 1:0. 5 were the best of all. The proper proportion between drugs and adjuvants in the tablet was 25 %. Through the results of in vitro release experiment proved different drug-release mechanism at different stage. Conclusion: The Qing-Huang-Zhu Buccat bioadhesive Tablets being prepared showed definite adhesive and sustained-release characteristics.
