摘要
通过DNA提取方法对比,采用CTAB/LiCl法提取甘薯黑斑病菌基因组DNA,再利用限制性内切酶BamH I对纯化的甘薯黑斑病菌基因组DNA进行部分酶切,获得大小为15kb左右的DNA片段。将经BamHI酶切消化的Lambda DASH II载体臂与DNA片段相连,然后以大肠杆菌XL1BlueMRA(P2)为寄主细胞,在国内首次成功构建了甘薯黑斑病菌(Ceratocystis fimbriata)基因组文库,所构建基因文库效价为4.12×105pfu/mL。
According to comparison of extraction methods, genomic DNA of Ceratocystis fimbriata was extracted by CTAB/LiCl method. While 15kb DNA fragments were obtained by partial digestion of purified genomic DNA of C. fimbr/ata with restriction enzyme BamH I. Repurified 15kb DNA fragments were ligated with Lambda DASH II Phage DNA vector arms digested by BamH I. And a genomic DNA library of C. fimbr/ata was constructed successfully with XL1BlueMRA(P2) as host cell. The titer of the genomic DNA library was 4.12× 10^5pfu/mL
