摘要 目的建立了HPLC-ELSD法测定注射用控喜树碱脂质体中杂质溶血磷脂酰胆碱的含量,并初步研究了影响脂质体类制剂中磷妇水解的因素。方法用NH2柱(150×4.6mm,5μm),以A:甲醇-氯仿(97:3),B:2%草酸.乙醇(16:84),A:B=7:3为流动相;柱温:30℃;流速为0.8mL·min^-1;进样量为20μL。结果注射用羟喜树碱脂质体中溶血磷脂酰胆碱能很好地分离检出。其线性浓度范围为:0.08~0.4mg·mL^-1(r=0.9991);加样回收率为98.25%,rds=1.6%。结论该方法简便。准确、分析时间短,适用于注射用羟喜树碱脂质体中磷脂有关物质的测定。 OBJECTIVE To determine the concentrations of lysophosphatidyl cholin in injectable HCPT liposome by HPLC method. METHODS A NH2 column (150mm × 4.6mm, 5μm) was used. The elution mobile phase was A: methanol-chloroform (97 : 3), B: 2 % oxalic acid-ethanol-( 16 : 84), A : B = 7 : 3 ; Tthe flow rate was 0.8mL· min^-1, and the detector was evaporative light-scattering detector (ELSD). RESULTS The linear range was0.08- 0.4mg· mL^-1 (r = 0.9991) and the average recovery rate was 98.25%, with the RSD 1.6% . CONCLUSION The method is rapid; accurate and reliable, and can be used to determine the lysophosphatidyl cholin in liposorne for injection.
