摘要 从灵芝菌丝体和子实体热水提取液中分离和纯化得到两种多糖,凝胶过滤层析测得分子量分别为3.7×104和4.2×104。气相色谱分析表明菌丝体多糖含D-葡萄糖、D-半乳糖、D-甘露糖、D-木糖、L-岩藻糖、L-鼠李糖,其摩尔比为5.35∶2.67∶1.00∶1.19∶0.38∶0.37;子实体多糖含D-葡萄糖、D-半乳糖、D-甘露糖、D-木糖、L-阿拉伯糖、L-鼠李糖,其摩尔比为5.82∶2.23∶1.00∶1.35∶0.72∶0.51。通过IR、13C-NMR、高碘酸盐氧化、甲基化分析、Smith降解、部分酸水解等手段对灵芝多糖的化学结构进行了研究,确定两种多糖都是以β(1→3)糖苷键为主链的杂多糖,在6-O和4-O上有分枝。 wo polysaccharides were isolated and purified from the hot water extract of the mycelia and fruiting body of Ganoderma lucidum(GL). Their mean molecular weights were estimated to be approximately 3.7×10 4 and 4.2×10 4 respectively. The components of these polysaccharides were identified by gas chromatography as:D glucose, D galactose, D mannose, D xylose, L fucose and L rhamnose in the molar ratio of 5.35∶2.67∶1.00∶1.19∶0.38∶0.37 in mycelia polysaccharide and D glucose, D galactose, D mannose, D xylose, L arabinose and L rhamnose in the molar ratio of 5.82∶2.23∶1.00∶1.35∶ 0.72 ∶0.51 in fruiting body polysaccharide. By means of IR, 13 C NMR,periodate oxidation, Smith degradation, methylation analysis and partial hydrolysis, etc, the structures of GLP have been identified, and it showed that both had a main chain of β (1→3)linked glucose residues with difference in the branching frequency and linkage mode of side chains.
