小鼠肾脏足细胞GPSD核心蛋白在高糖环境下表达的变化

摘要 目的通过观察高糖环境培养的肾小球足细胞裂孔隔膜(GPSD)核心蛋白nephrin、podocin和CD2AP的表达,探讨糖尿病肾病蛋白尿的发生机制。方法以RT—PCR方法检测不同浓度(5、10、20和40mmol/L)葡萄糖培养液培养小鼠足细胞24h后nephrin、podocin和CD2AP mRNA的表达水平。结果足细胞nephrin mRNA的表达在5mmol/L葡萄糖培养液培养24h后即显著减弱(P〈0.05),在其它浓度葡萄糖培养液培养24h后几乎无表达(均P〈0.01);podocin和CD2AP mRNA的表达在10mmol/L葡萄糖培养液培养24h后显著下降(P〈0.01)。但随着培养液的葡萄糖浓度增加,podocin和CD2AP mRNA的表达却逐渐上调。结论口糖环境抑制足细胞nephrin、podocin和CD2AP的表达,可能是糖尿病肾病蛋白尿发生、发展机制之一。 Objective To investigate the effect of high glucose on the expression of glomeroular podocyte slide diaphrame (GPSD) core proteins (nephrin, podocin and CD2AP) in mouse podocytes. Methods Mouse podocytes were cultured in medium with glucose concentration of 0 (control), 5, 10, 20 and 40 mmol/L for 24 hrs. The expressions of nephrin, podocin and CD2AP mRNA were examined by RT-PCR. Results Compared with the control group, the expression of nephrin mRNA in 5 mmol/L glucose group significantly decreased (P 〈 0.05), while the expression in 10, 20 and 40 mmol/L glucose groups was not detected. The expressions of podocin and CD2AP mRNA in 10 mmol/L glucose group were significantly decreased in comparison with those of control group (P 〈0.01 and 0.05), while the expressions of podocin and CD2AP mRNA increased gradually as the concentration of glucose increased from 10 mmol/L to 40 mmol/L. Conclusion The expressions of mouse podocyte nephrin, podocin and CD2AP are inhibited in high glucose environment, which may be associated with the development of proteinuria in diabetic nephritis.
