摘要
目的研究凋亡相关基因B细胞淋巴瘤/白血病-2及其同源蛋白(Bcl-2/Bax)及肿瘤坏死因子和神经生长因子受体跨膜蛋白及其配体(Fas/Fasl)在子宫内膜息肉中的表达,探讨子宫内膜息肉形成的原因和机制。方法选择子宫内膜息肉患者63例作为研究组,正常子宫内膜者55例作为对照组,采用免疫组织化学二步法检测两组患者增生期及分泌期Bcl-2、Bax及Fas、Fasl的表达,作半定量分析后进行组间比较。结果(1)与对照组增生期比较,研究组增生期Bcl-2及Bcl-2/Bax均明显升高(均P〈0.05);与对照组分泌期比较,研究组分泌期Bcl-2、Bcl-2/Bax均明显升高(均P〈0.05),而Bax明显降低(P〈0.05)。(2)研究组和对照组增生期Fas均呈中等强度表达,而且研究组增生期Fas的表达明显低于对照组(P〈0.05);研究组分泌期Fas及Fasl的表达减弱,均明显低于对照组(均P〈0.05)。(3)研究组Bcl-2的表达与Fas的表达无明显相关性(r=-0.083,P〉0.05)。而对照组Bcl-2的表达与Fas的表达呈显著负相关(r=-0.934,P〈0.01)。结论凋亡机制调控失常可能参与了子宫内膜息肉的发生、发展。
Objective To investigate the expression of B cell lymphoma/leukemia-2 and its homologous protein (Bcl-2/Bax), tumor necrosis factor and nerve growth factor receptor transmembrane protein and its ligand (Fas/Fasl) In endometrial polyps (EMPs). Methods Bcl-2, Bax, Fas and Fast were detected using immunohistochemistry method (EnVision system) in 63 EMP tissue specimens (study group) and 55 normal endometrium specimens (control group). Semi-quantitative analysis was used to assess the average value of staining. Results Bcl-2 expression in EMPs of the proliferative phase was higher than that in normal endometrium (P〈0.05), Bax expression in EMPs of secretory phase was lower than that in normal endometrium (P〈0.05). Fas expression in EMPs of proliferative phase was lower than that in normal endometrium (P〈0.05), Fasl expression in the EMPs of secretory phase was lower than that in normal endometrium (P〈0.05). In the EMP there was no significant correlation between Bcl-2 expression and Fas expression (r=-0.083, P=0.174). But in the normal endometrium Bcl-2 expression was negatively correlated with Fas expression (r=-0.934, P〈0.01). Conclusion The defected regulation of apoptosis may play a role in the pathogenesis of EMP.
