摘要
目的探讨重症急性胰腺炎(SAP)小鼠肺组织粘附分子(ICAM-1、P-selectin、E-selectin)的表达及其与肺损伤的关系。方法将40只ICR雌性小鼠随机分为SAP组和对照组。SAP组采用连续7次(每次间隔1h)腹腔内注射雨蛙素及脂多糖制作小鼠SAP肺损伤模型,对照组用0.9%氯化钠注射液。最后一次注射后2h麻醉小鼠,自右颈静脉取血检测血淀粉酶(Amy)、乳酸脱氢酶(LDH)、C-反应蛋白(CRP);光镜下观察小鼠肺脏的病理形态,测定肺组织中髓过氧化物酶(MPO)水平。PCR法测定肺组织中粘附分子的含量。结果SAP组小鼠的Amy、LDH、CRP明显升高(P〈0.05);肺脏出现明显的病理损害;肺组织中MPO水平、粘附分子的含量均同步上升(P〈0.01)。结论SAP小鼠肺组织中粘附分子(ICAM-1、P-selectin、E-selectin)过度表达,介导了中性粒细胞在肺组织中的浸润,在肺损伤发生、发展过程中起重要作用。
Objective To evaluate the intrapulmonary expression of ICAM-1, P-selectin and E-selectin and its relationship with acute lung injury in severe acute pancreatitis. Methods 40 female mices were divided in SAP group and control group (n=10). Severe acute pancreatitis was induced by cerulin and lipopolysaccharide (LPS) in mice. The expression of Amy, LDH and CRP in serum and activity of myeloperoxidase (MPO) in lung were evaluated and mRNA of intercellular adhension molecule 1, P-selectin, E-selectin in lung injury were assessed by real-time reversetranscriptase-polymerase chain reaction (RT-PCR). Pathological changes was also observed. Results In comparison with control group, The expression of Amy,LDH and CRP in serum were increased, pathological changes of lung tissue were aggraved significantly, mRNA of intercellular adhension molecule 1, P-selectin, E-selectin were synchronously evaluated in lung tissue as the activity of MPO. Conclusion The excess expression of intercellular adhension molecule 1, P-selectin, E-selectin, leading to infiltration of neutrophile in the lung, had a great effect on acute pancreatitis associated lung injury.
