白血病干细胞与SDF-1/CXCR4生物学轴关系的研究进展

摘要 急性髓细胞性白血病是造血干细胞克隆性增殖的肿瘤性疾病,其发病机制至今仍不十分清楚,而且髓外浸润和侵袭仍是导致复发和病人死亡的重要原因之一。因此,有关急性白血病发生、发展和髓外浸润的机制和相应的治疗学研究成为当今血液学研究的重点和难点之一。已有的研究表明,基质细胞衍生因子1(SDF-1)及其特异性受体CXCR4所组成的SDF-1/CXCR4轴可通过改变白血病骨髓造血微环境,在白血病细胞的增殖、耐药、浸润中发挥一定的作用。本文重点探讨SDF-1/CXCR4轴在白血病干细胞迁移与归巢中的作用。 Acute myeloid leukemia is a malignant disease from the hemopoietic stem cell( HSC), whose nosogenesis were still clear. Furthermore, the infiltration and invasion of leukemia cells outside the marrow are still the most important reasons for recurrence and the death of patients. 33nerefore ,it becomes the emphasis for leukemia to approach the mechanism of its genesis, development and the invasion outside the marrow, and the research for its therapeuties is even more important. The SDF-1/ CXCR4 axis, which consists of stromal cell derived factor-1 and its only receptor CXCR4, may play an important role in the proliferation and drug resistance of the leukemia cells via its function of changing the mieroenvironment of bone marrow. This article reviewed the function of SDF-1/CXCR4 axis in migration and homing of leukemia stem cells.
