摘要
目的探讨阿司匹林对急性肺损伤大鼠肺组织血红素加氧酶-1表达的影响。方法24只健康雄性SD大鼠,体质量180~220g,随机分为三组(n:8),正常对照组(A组)、油酸组(B组)及油酸阿司匹林组(C组)。三组大鼠腹腔注射10%的水合氯醛(0.35mL/100g)麻醉,麻醉生效后,B、C组分别股静脉注射油酸(0.15mL/kg)建立大鼠ALI模型,正常组注射等量生理盐水。于油酸注射后2h时,行血气分析,计算氧合指数(PaO2/FiO2);称量肺组织湿质量(W)、干质量(D),计算肺组织湿干质量比(W/D);光镜观察肺组织病理性改变;采用实时荧光定量PCR测定肺组织HO-1mRNA的表达水平。结果PaO2/FiO2:与A组比较,B、C组显著降低(P〈0.05),C组较B组显著升高(P〈0.05)。W/D:与A组比较,B、c组显著升高(P〈0.01),C组较B组显著降低(P〈0.01)。HO-1mRNA表达:与A组比较,B、C组显著升高(P〈0.01),C组较B组显著升高(P〈0.01)。光镜下肺组织病理改变:B组可见肺组织水肿,炎性细胞浸润,肺泡壁增厚,肺泡隔增宽,肺泡腔内大量渗液,毛细血管内微血栓形成和自细胞附壁、聚集、血液淤滞,并有局灶性肺不张和肺气肿存在。C组亦可见肺组织水肿,炎性细胞浸润,肺泡间隔增宽,白细胞附壁、聚集、血液淤滞等现象,但程度较B组明显减轻。A组则未见出血、水肿、肺泡腔内渗液和毛细胞血管内微血栓、炎性细胞附壁、聚集等现象。结论阿司匹林可通过增加肺组织中血红素加氧酶-1的表达而发挥保护作用。
Objective To explore the effect of aspirin on the expression of hemeoxygenase - 1 ( HO - 11 in acute lung injury in rats. Methods 24 healthy male SD rats, each weighting 180 ~ 220 g, were randomly divided into three groups ( n =8 ) : group A, group B, group C. All the rats were anesthesitized with 10% chloral hydrate (0. 35 mL/lOOg). Group A were given O. 15 mL/kg natural solution through the femoral vein;group B, C were given 0. 15 mL/kg oleic acid through the femoral vein. At 2 h after intravenous oleie acid, blood sample were take from the carotid artery for blood gas analysis, and PaO2/FiO2 was calculated ;the left lung tissues were collected in 2 h after OA for lung weight/dry ratio (W/D) ; specimens of lung tissue were observed for the pathological change of lung structure with optical microscope, RT - PCR methods were used to determine the expression level of HO - 1 mRNA in lung tissue. Results PaO2/FiO2 :the level of PaO2/FiO2 was significantly lower in group B, C than thatin group A ( P 〈0. 05 ). The level of PaO2/FiO2 was significantly increased in group C than that in group B ( P 〈 0. 05 ). W/D : the level of W/D was significantly increased in group B, C than that in group A ( P 〈0. 01 ) ,the level of W/D was significantly lower in group C than that in group B ( P 〈0. 01 ). HO - 1 mRNA: the level of HO - 1 mRNA in the lung tissue was significantly increased in group B, C than that in group A ( P 〈0.01 ) , the level of HO - 1 mRNA in the lung tissue was significantly increased in group C than that in group B ( P 〈 0. 01 ). In group B, The structure of lung tissue in optical microscope was of very disorder, massive inflammatory cell infiltrating, alveolus capillary dilating, congestioning, alveolar wall congestioning obviously, alveolar septum widening, even massive erythrocyte appearing in alveolar space, focal atelectasis. The injury of lung tissue was significantly abated in group C than that in group B. Conclusion Aspirin may protect the rats from acute lung injury through increasing the expression of HO - 1.
