摘要 研究了薄层扫描法测定万寿菊中叶黄素含量的方法。确定以硅胶G薄层板为固定相,石油醚-氯仿-丙酮(12∶5∶4)为展开剂,扫描波长445 nm,参比波长700 nm,采用反射法锯齿扫描;万寿菊中叶黄素酯的提取与皂化条件为:正己烷-丙酮-无水乙醇-甲苯(10∶7∶6∶7)为提取剂,料液比1∶80,10%KOH-CH3OH溶液为皂化液,皂化温度为45℃,皂化时间为40 min,水浴后暗处静置80 min,再加8 mL正己烷并用10%Na2SO4溶液定容,振荡后静置30 min。实验表明,在0.336~2.016μg范围内,叶黄素浓度与扫描峰面积呈良好的线性关系,该方法具有良好的精密性和稳定性,可用于分析万寿菊中叶黄素的含量。 The TLC method was used to determine lutein. The zigzag scanning method was adopted. The fixed phase was silica gel(G) plate, the developing agent was ligroin-chloroform-acetone(volume ratio 12 : 5 : 4);the scanning wavelength was 445 nm and the reference wavelength was 700 nm. The lutein was exeracted from marigold flower by organic solvent. The extracting agent was a mixture of n-hexaneacetone-absolute ethyl alcohol-methylbenzene (volume ratio 10 : 7 : 6 : 7), the ratio of extracting agent was 1 : 80(g/mL) ,and the saponification agent was 10% KOH-methanol. The solution was kept in water bath for 40 min of 45 ℃ ,and then placed in the dark for 80 min. 8 mL of n-hexane and 10% Na2SO4 were added into the solution, and then it was kept still for 30 min after shaking. The result showed that the linearity of lutein was in the range of 0. 056-0.336μg/mL. The TLC method was reliable to determination the lutein in marigold flowers and the method was accurate and reproducille.
