摘要 建立了LC-MS/MS法测定大鼠血浆中的咪达唑仑。血浆样品经碱化处理后再用乙醚提取。采用C_(18)色谱柱,以乙腈-0.1%甲酸(60:40)为流动相,三唑仑为内标,采用电喷雾电离源(ESI),选择性正离子多离子反应监测(MRM)m/z 326.0→291.0(咪达唑仑)和m/z 343.0→308.0(三唑仑)。咪达唑仑在1~500 ng/ml浓度范围内线性关系良好。方法回收率大于95%,提取回收率大于76%,日内和日间RSD小于8%。 A LC-MS/MS method was established for the determination of midazolam in rat plasma. The drug was extracted from plasma samples with ethyl ether after alkalization. A C18 column was used with the mobile phase of acetonitrile-0.1% formic acid (60 : 40). Triazolam was used as internal standard. ESI was applied in positive ion mode. The detected ions by multiple reaction monitoring (MRM) mode were m/z 326.0→291.0 (midazolam) and m/z 343.0→308.0 (triazolam). The calibration curve was linear in the concentration range of 1 - 500 ng/ml. The average method recovery was over 95 % and the average extraction recovery was above 76 %. The inter- and intra-day RSDs were less than 8 %.
