摘要
以控制绵羊高繁殖力的BMPR-IB、BMP15和GDF9基因为候选基因,采用PCR-RFLP方法分析滩羊BMPR-IB,BMP15和GDF9基因多态性与繁殖性状的关系。试验结果表明,滩羊的BMPR-IB基因出现了两个SNP位点,且在相应位置发生了与Booroola羊相同的突变(A746G),该基因++基因型在滩羊群体内是优势基因型,但是双胎母羊和其后代的BB和B+基因型频率均高于单胎母羊,双胎羔羊和单胎母羊++和B+基因型频率最小二乘均值差异显著(P<0.05),两个母羊群体各基因型之间平均产羔数差异不显著(P>0.05),从而推测含有BMPR-IB基因突变的滩羊具有产双羔的潜力,可以作为滩羊选种选育的一项指标。而BMP15基因的B4突变(G→T)和GDF9基因的G8突变(C→T)在滩羊上不表现多态性,因此排除了BMP15基因的B4突变和GDF9基因的G8突变是影响滩羊繁殖性能的可能性。
BMPR-IB ,BMP15 and GDF9 which control the fecundity of sheep were studied as candi- date genes. PCR-RFLP was applied to analyze the relationship between the gene and genotype poly- morphisms of BMPR-IB ,BMP15 and GDF9 in Tan sheep and their prolific trait. The results showed that there were two SNP mutation and had a same mutation in BMPR-IB gene (A746G) of Tan sheep as that of Booroola. The ++ mutation genotype was superior in Tan sheep, but the double fetal ewe and filial with BB and B+ genotype frequency more than single fetal ewe. The difference of the least squares means (LSM) for the average litter size of every mutation genotype was non-significant (P) 0.05) in two ewe groups. It could be inferred that the Tan sheep had product twin potential of BMPR-IB gene mutation and as a index of selection. In addition, B4 mutation(G→T) of BMP15 gene and G8 mutation(C→T)of GDP9 gene were not detected in Tan sheep. Then it was ruled out the possibility that the fecundity of Tan sheep was affected by the B4 mutation of BMP15 and G8 mutation of GDP9 gene.
