摘要 为了探讨Ghrelin在鸵鸟小肠发育过程的表达特点及其调节食欲的细胞学基础,采用免疫组化SP法分别检测了3日龄、3周龄和9~11月龄鸵鸟小肠内Ghrelin阳性产物的分布及变化特点。结果显示:Ghrelin免疫阳性产物在鸵鸟3个年龄组各段小肠内均有分布,主要分布在小肠的黏膜层,其中固有层的小肠腺和肠绒毛上皮分布较多,包括柱状细胞,内分泌细胞和淋巴细胞等;阳性产物的表达量随年龄增加呈递增趋势,3日龄鸵鸟阳性细胞相对表达量最少,3周龄鸵鸟次之,9~11月龄最多,且差异显著(P〈0.05);不同肠段Ghretin表达量亦有不同,但各年龄段均表现为自十二指肠、空肠、回肠阳性产物相对表达量依次降低,其中3日龄和3周龄鸵鸟各肠段阳性细胞相对表达量变化差异不显著(P〉0.05);9~11月龄鸵鸟各肠段阳性细胞相对表达量变化差异显著(P〈0.05)。表明小肠中Ghretin的表达随年龄的增长而增多,并通过自分泌兼或旁分泌的形式调节小肠消化功能的发育和完善。 To study the the age-related expression rules of Ghrelin in small intestine of ostrich and the foundation of cytology on regulation of appetite with Ghrelin. The expression and changes of ghrelinimunopositive (ghrelin-ip) cell were detected with immunohistochemical SP method in Ostrich's small intestine during 3 days, 3 weeks and 9~11 months. Results: Ostrich's small intestine of different ages all had ghrelin-ip cell , it was distributed in Mucosal layer of small intestine , there were more ghrelinip cells in the intestinal villus and the glands of small intestine than that in other sections of it. Ghre- lin-ip cell was columnar cell, endocrine cell, lymphocyte and so on; The expression of ghrelin-ip cell increased with growing. Compared with the 3 weeks of ostrich ,the expression of 9~11 months of ostrich increased obviously ,and the 3 days of ostrich had the least expression; The expression of Ghrelin-ip cells also was different in duodenum, jejunum and ileum. The duodenum had the most of expression,the ileum had the least of that. The difference of relative expression of ghrelin-ip cell in every intestine during 3 days and 3 weeks was not conspicuous( P〉0.05),but it during 9~11 months was conspicuous(P〈0.05). It manifested that the expression of Ghrelin increased with growing. Ghrelin might regulate the development of small intestine through autoeine and paracrine.
