摘要
应用猪圆环病毒Ⅱ型(Porcine circovirus 2,PCV2)ORF2重组蛋白,研制诊断PCV2的抗体试剂。利用原核表达系统表达PCV2 ORF2蛋白,经12%浓度的SDS-PAGE凝胶电泳分析后免疫家兔制备抗PCV2ORF2重组蛋白抗体,并用间接酶联免疫吸附试验(indirect-ELISA)确定其效价,然后以Western blotting法分析该抗体特异性。结果表明:PCV2 ORF2重组蛋白被表达,且在免疫家兔后获得效价高、特异性好的抗体。兔抗PCV2-ORF2重组蛋白抗体以其特异性好的特点可以作为免疫诊断试剂。
PCV2-ORF2 recombinant protein was used and prepared immunodiagnosis antibody agent for PCV2. The structural protein encoded by PCV2-ORF2 was expressed in recombinant E. col BL21 （DE3） and inclusion body analyzed by 12% SDS-PAGE then immunized rabbits to prepare PCV2- ORF2 recombinant protein antibody and its titer was tested by indirect-ELISA then the antibody spe- cificity analyzed by Western blotting. The result showed PCV2-ORF2 recombinant protein product expressed then obtained highest titer and fine specificity antibody by immunized rabbits. Antibody of rabbits against PCV2-ORF2 recombinant protein could be made immunodiagnosis reagent for its fine specificity.
