菜用甘薯遗传多样性的ISSR分析 被引量:12

摘要 利用ISSR分子标记分析了10个菜用甘薯材料的遗传多样性和亲缘关系。结果表明:11个ISSR引物在供试材料中扩增出83个清晰条带,其中76个具有多态性,多态性比率为91.57%,各扩增条带的多态性信息指数(PIC)平均为0.247。种质间遗传相似系数变化范围为0.578~0.904,平均值为0.645,显示菜用甘薯材料的遗传差异较大。通过聚类分析将10份菜用甘薯种质分为三个类群,其中两份广东材料与台农71聚为一类,且遗传相似性较高,福建的两份材料与徐州的两份材料及广东的一份材料聚为一类,而河南的商薯19与广东的另一份材料聚为独立的一类,他们与其他两类的亲缘关系较远。本研究的结果为菜用甘薯杂交育种的亲本选配提供了理论依据。 In order to provide theoretical information for vine-vegetable sweet potato breeding, this study was to probe the relationship and genetic diversity of the sweet potato germplasms. Ten vinevegetable sweet potato species were used to detect the intra-and inter-specific genetic variation by inter-simple sequence repeat (ISSR) analysis. 87 DNA fragments were amplified, using 11 primers screened from 60 ISSR primers, among which 83 DNA bands were polymorphic(PPB= 91.57 %). The average polymorphism information content (PIC) was 0. 247, which indicated that genetic basis was relatively spacious. The cluster analysis based on ISSR markers revealed that the two cultivars from Guangdong and Tainong 71 was clustered into a group. Two cultivars from Fujian and two cultivars from Jiangsu was clustered into a group; A cultivar from Henan and a cultivar from Guangdong was clustered into another group. The genetic relationship of all 10 vine-vegetable sweet potato cuhivars revealed by the ISSR dendrogram would be of great help to accurately select the parents in breeding programs.
