摘要
目的:观察三七总皂苷含药血清对溶血磷脂酸(LPA)诱导的血管平滑肌细胞增殖效应的影响,分析该作用与细胞外信号调节激酶ERK信号通路的关系。方法:采用贴块法培养大鼠主动脉平滑肌细胞;以四甲基偶氮唑蓝(MTT)比色法观察不同分组对细胞增殖的影响;采用流式细胞仪分析细胞周期和DNA含量。结果:空白血清+LPA组较空白血清组OD值升高(P<0.05),S期指数升高(P<0.01);三七总皂苷含药血清+LPA组与卡托普利含药血清+LPA组比较OD值降低(P<0.05),S期指数无差异;三七总皂苷含药血清+LPA+PD98059组较空白血清组OD值降低(P<0.05),S期指数无差异。结论:三七总皂苷对LPA诱导的VSMC增殖有拮抗作用,且作用强于卡托普利,其作用机制可能与抑制LPA对ERK信号通路的激活,从而抑制DNA合成,阻止细胞进入S期有关。
Objective : To investigate the effect of the serum containing panax notoginseng saponins on the proliferation of vascular smooth muscle cells(VSMC) induced by Lysophosphatidie Acid(LPA) and analyze the relationship with ERK signal pathway. Methods:Rat VSMCs were obtainded by tissue - piece inoculation; effects of different groups on the proliferation of vascular smooth muscle cells was evaluated by MTT colorimetry ; analyses of cell cycle and DNA content were performed by flow cytometry. Results : The optical density(OD) and the S phase fraction(SPF) of the LPA + blank serum group were increased than the blank serum group(P 〈0. 05 ,P 〈0. 01 ) ;In comparison with the LPA + captopril - containing serum group,the OD of the LPA + panax notoginseng saponins- containing serum group was decreased( P 〈 0. 05 ), SPF has no difference;the OD of the PD98059 + LPA + panax notoginseng saponins -containing serum group was decreased( P 〈 0. 05 ), SPF has no difference compared to the blank serum group. Conclusion:Panax notoginseng saponins can inhibite the proliferation induced by LPA, and the effect is greater than eaptopril . The mechanism for such effect may be associated with the block of ERK signal pathway actived by LPA,inhibition of DNA synthesis and preventing the transition from G0/G1 into S phase of VSMCs
