细胞冻存条件对单细胞凝胶电泳试验结果的影响

摘要 将未处理的(对照)和重铬酸钾处理的V79细胞贮存于4℃、-20℃、-80℃和-196℃,于保存后的0,4,24,48,168h取出进行单细胞凝胶电泳(SCGE)试验。结果显示各温度保存24h的细胞DNA迁移长度与新鲜细胞无差异;更长时间(48和168h)的保存细胞DNA迁移长度明显增加;同时各温度保存的细胞存活率均随贮存时间的延长而下降。因此,24h内细胞保存以4℃为好。 V 79 cells treated with potassium dichromate and control cells were stored at 4℃, -20℃,-80℃ and -196℃. Single cell gel electrophoresis (SCGE) was carried out after preserving cells for 0, 4, 24, 48 and 168 hours. The results showed that there was no difference on DNA migration between cells processing immediately after trypsinization and cells preserving for 24h at different low temperatures. Longer storage (48h or 168h) resulted in a significant increase in the length of DNA migration and decrease in cell viability. The results suggested that 4℃ is more convenient for less than 24h storage without influence on DNA migration and -80℃ is better for longer storage to minimize the influence on DNA migration.
