摘要
目的研究小鼠透明带溶解液诱导人精子顶体反应的效果,建立人精子顶体反应模型。方法正常生育力人精子经Biggers,WhitenandWhitinghammedium(BWW)获能5~6h后,分别用小鼠透明带溶解液(mZPS)、mZPS的缓冲液halogenbufer(HB)、Ca2+载体A23187处理1h;对实验和对照组精子用经典的豌豆凝集素(PSA)法检测顶体反应。结果mZPS组的顶体反应率高于对照组。结论mZPS能诱导人精子顶体反应,透明带诱导精子顶体反应具有一定的种间交叉性。
Objective\ To investigate the induction of acrosome reaction in human spermatozoa by solubilized mouse zona pellucida(mZPS). \ Methods\ The swimmingup spermatozoa were incubated in Biggers,Whitten and Whittingham medium(BWW) for 5~6 hours at 37℃,and then treated,with mZPS,halogen buffer(the buffer of mZPS),or wellknown acrosome reaction activators calcium ionophose A23187 respectively.\ After an hour.the percentages of acrosomereacted spermatozoa identified by fluoresceinated pisumsativum agglutinin(PSA)and visuallized by fluorescence microscopy.\ Results\ The acrosome reaction rate after spermatozoa treated with mZPS for 1 hour was higher than negative controls. \ Concluslon\ Solubilized mouse zona pellucida are capable of inducing acrosome reaction in human spermatozoa.
