摘要
目的:探讨肾癌组织中WNT诱导的分泌型蛋白-1(WISP-1)基因的表达及意义。方法:应用实时RT-PCR和免疫组化法检测31例肾癌组织和18例癌旁正常肾组织中WISP-1mRNA和蛋白的表达。结果:WISP-1mRNA在肾癌组织中的表达中位值(3.37)显著低于癌旁正常组织(9.32),P<0.05;乳头状肾癌(4.67)显著高于透明细胞癌(2.20),P<0.05。WISP-1表达水平与肿瘤分期、大小无显著相关。肾癌组织和正常肾组织中WISP-1免疫组化阳性率分别为71.0%和100%,P<0.05。结论:WISP-1可能与肾癌的发生和分化相关。
OBJECTIVE: To investigate the expression of Wnt-in- duced secreted protein-1 (WISP-l) in human renal cell carcinoma (RCC) and determine whether it is correlated with RCC. METHODS: Thirty-one samples of RCC and 18 normal kidney tissues were collected. WISP-1 mRNA was detected by relative quantitative real-time RT-PCR and WISP-1 protein was examined by immunohistochemical staining. RESULTS: The media level of WISP-1 mRNA decreased sig- nificantly in tumor (3.37) compared with normal kidney tissues (9. 32, P〈0.05). The WISP-1 mRNA median level was higher in papillary RCCs (4.67) than in clear cell RCCs (2.20, P〈0.05). WISP-1 mRNA expression had no relation with the stage, grade and size of RCCs. The positive rate of immunohistochemical staining was also lower in RCCs (71.0%) than in normal kidney tissues (100%, P〈0. 05). CONCLUSION: WISP-1 may be correlated with the carcinogenesis and progression of RCC.
