摘要 目的:探讨核心蛋白聚糖(decorin,DCN)对HuH7细胞系体外增殖的影响及机制。方法:加入不同浓度(0、25、50、75、100、125、150、200μg/L)的DCN培养HuH7细胞系不同时间(12、24、48、72h和2周),用二甲基噻唑二苯基四唑溴盐(MTT)法及克隆实验测定细胞增殖速度和活力,流式细胞术测定细胞周期和凋亡情况。结果:DCN浓度增加及作用时间延长,细胞增殖速度减慢、活力减低,G1期细胞增多,凋亡增加。结论:DCN可能为一种负性调控蛋白,可通过调节细胞周期,抑制肝癌细胞的增殖,促进其凋亡。 AIM:To investigate the effects and mechanism of decorin( DCN) on the proliferation of HuH7 hepatoma carcinoma cell line in vitro. METHODS:Hepatoma carcinoma cells was cultured with DCN in different concentration (0,25,50,75,100,125,150,200 μg/L) for different time(12,24,48,72 h and 2 weeks). Cell activities were studied by MTT and clone test. The changes of cell cycle and apoptosis were analyzed by Flow cytometry. RESULTS:The proliferation of HuH7 cells could be inhibited by DCN in vitro and the inhibition effect was the time and dose dependent relationship. DCN could block cell cycle at G1 phase. Apoptosis of hepatocarcinoma cells could be effi ciently induced by DCN in a time/dose-dependent manner. CONCLUSION: DCN may be a negative regulatory protein inhibiting hepatoma carcinoma cell proliferation through in- hibiting cell cycle and inducing apoptosis of cell in vitro.
