抗CD3-抗B细胞白血病独特型双特异性抗体的制备及细胞毒性

Preparation and cytotoxicity of anti CD3×Anti B CLL idiotype bispecific antibody

采用化学偶联法制备抗ＣＤ３抗Ｂ细胞性白血病独特型双特异性抗体，研究了该双特异性抗体诱导ＬＡＫ细胞的增殖及其细胞毒作用。结果提示，双特异性抗体可显著提高ＬＡＫ细胞与Ｄａｕｄｉ细胞的结合率；促进淋巴细胞的增殖。乳酸脱氢酶试验检测发现，加入双特异性抗体后，ＬＡＫ细胞对Ｄａｕｄｉ细胞的细胞毒性作用显著高于对照组。

The anti CD3×anti B CLL idiotype bispecific antibody(BsAb) was prepared by chemical conjugation. The effect of BsAb on the proliferation and cytotoxicity of LAK cells were studied. The cytotoxic activity of LAK cells was determined by LDH assay. The results showed that BsAb significantly increased the conjugate formation between LAK and Daudi cells and enhanced proliferation of lymphocytes and could enhance effects on the cytotoxicity against Daudi cells.
