摘要
以不同季节的苦楝枝条为研究对象,利用光学显微技术和SDS-PAGE技术研究其枝条中贮藏蛋白在休眠期和生长期的分布情况。经汞-溴酚蓝染色后的石蜡切片显微观察表明,休眠期枝条细胞的液泡中存在大量蓝色近椭圆状蛋白颗粒,而在生长期枝条细胞的液泡中则未发现这种蛋白颗粒的存在。SDS-PAGE结果表明:休眠期枝条中的蛋白条带数目明显多于生长期枝条且浓度高,有123、03、23、84、05、3kD 6条蛋白带在休眠期枝条细胞中大量特异表达。
Vegetative storage proteins(VSPs)in the Melia azedarach L.were identified by using optical microscopy and SDS-PAGE.Under optical microscope,deep blue masses in a granular appearance were observed in the secondary xylem parenchyma cells of the dormancy season barks after stained with mercury-bromophenol blue.However,there was no granular protein in the development season barks.Compared with SDS-PAGE results of the development season barks,there were six 12,30,32,38,40,53 kD major proteins expressed in the dormancy season barks,which was probably the vegetative storage protein of Melia azedarach L.
