人外周血γδT细胞对肺癌细胞杀伤作用的研究 被引量:2

摘要 目的:探讨人外周血扩增的γδT细胞对肺腺癌细胞A549的杀伤作用。方法:取健康人外周血分离单个核细胞,置于RPMI1640培养基(含100ml/L小牛血清,50ml/L人AB型血清,IPP2μg/L,IL-2105IU/L)中培养扩增γδT细胞,用流式细胞仪检测γδT细胞的纯度。用扩增后的γδT细胞与肺腺癌细胞A549按不同效靶比例孵育,用乳酸脱氢酶释放法进行细胞毒活性测定。结果:人外周血单个核细胞经过培养扩增γδT细胞可以达到较高的纯度。人外周血γδT细胞对人肺腺癌细胞A549有较强的杀伤活性,其杀伤活性与效靶比和效应细胞的纯度呈正相关。人外周血γδT细胞对靶细胞的杀伤活性要高于CIK细胞。结论:人外周血扩增的γδT细胞对肺腺癌细胞A549有较强的杀伤作用。 Objective:To investigate the cytotoxic effects of human peripheral blood γδt Cells on lung cancer cell lines A549. Methods:The γδt Cells in peripheral blood of healthy volunteers were cultured using interleukin-2(IL-2)and isopentenyl pyrophodphate (IPP)in PRMI1640 medium. The purity of γδT Cells was analyzed by flow cytometry .The cytotoxic activity of the purified γδT Cells against human lung cancer cell lines A549 was tested in different effector-to-target ratios (T/E ratios). Results:γδT Cells were obtained with a high purity from peripheral blood mononuclear cells,the cytotoxic effects of γδT Cells on A549 depended on γδT cells' purity and effector-to-target ratios. When effector-to-target ratios and purity of γδT cells increased,the cytotoxcity effect on A549 increased. γδT Cells showed more remarkable cytolysis than CIK cells. Conclusion:γδT Cells of human peripheral blood could significantly kill lung cancer cell lines A549 in vitro.
