摘要 【目的】研究双酚A(BPA)对小鼠。肾组织的毒性效应。【方法】将BPA溶于金龙食用油中,备用。将20只小鼠随机分为5组:I组为对照组,注射食用油1mL/只;Ⅱ~V组为药物处理组,分别注射0.1,0.5,1或2mg/mL的BPA1mL/只,每5d注射1次,连续注射3次。于末次注射后第5,10,15和20天各组分别取1只小鼠处死,取肾脏,用多聚甲醛固定,常规方法制备切片,免疫组织化学染色显微观察pactin的分布定位,分析BPA对小鼠肾组织β—actin阳性细胞表达的影响。【结果】第5~15天,0.1和0.5mg/mLBPA处理组小鼠肾组织β-actin阳性细胞数量与对照组差异不显著,1和2mg/mL BPA处理组小鼠肾组织阳性细胞数量与其他3组差异显著;第20天,各BPA处理组小鼠β—actin阳性细胞数量与对照组差异显著。【结论】小鼠肾组织中β—actin阳性细胞随BPA剂量的增加与用药时间的延长而减少。 [Objective] The toxicity of intraperitoneal injection of bisphenol A (BPA) of different doses to kidney tissue of mice was evaluated with the change of β-aetin expression. [Method] BPA was resolved in Jinlong plant oil. 20 mice were divided into 5 groups. One group injected with the plant oil was used as control. The other groups were injected with different doses of BPA (0.1,0.5,1 or 2 mg/mL per mice) at intervals of 5 d for 3 times respectively. After the injection of BPA or oil for 5,10,15 and 20 d,the mice were killed respectively. Then,each mouse kidney tissue was removed and fixed in polyformaldehyde. The change of β-aetin expression kidney tissue was observed by immunohistochemical staining. [Result] Compared with control group, after treatment with BPA from 5 d to 15 d,β-actin expression showed no change with the lower dose of BPA at 0.1 and 0.5 μg/mL. However,it was decreased significantly with the higher dose of BPA at 1 and 2 mg/mL. After the treatment with BPA for 20 d, the β-actin expression in all of groups treated with BPA was reduced greatly. [Conclusion] These results indicated that the number of β-actin-stained positive kidney cells was reduced by bisphenol A at a dose-dependent and time-dependent manner,and thus the systematic bisphenol A showed the strong toxicity of mice kidney tissue.
