丹参对新生大鼠高氧性肺损伤细胞因子表达的影响

Effects of red sage root on the expression of cytokines in hyperoxia-induced lung injury in neonatal rats

摘要 目的探讨丹参对新生大鼠高氧性肺损伤细胞因子表达的影响。方法出生2d的SD大鼠随机分为空气加生理盐水组(Ⅰ组)、空气加丹参组(Ⅱ组)、高氧加生理盐水组(Ⅲ组)和高氧加丹参组(Ⅳ组)。生后14d称重,记录体质量和肺质量,实时荧光定量PCR检测肺组织中白细胞介素6(IL-6),单核细胞趋化蛋白1(MCP-1),中性粒细胞趋化因子1(CINC-1)和双调蛋白(AR)基因相对水平。结果Ⅲ组体质量、肺质量、肺质量/体质量、肺湿重/肺干重低于Ⅰ组(P<0.05),Ⅳ组体质量、肺质量、肺质量/体质量、肺湿重/肺干重高于Ⅲ组(P<0.05)。Ⅲ组细胞因子IL-6、AR、MCP-1、CINC-1基因表达量明显高于Ⅰ组,Ⅳ组IL-6、AR、MCP-1、CINC-1基因表达量较Ⅱ组增加,但小于Ⅰ组(P<0.05)。结论丹参能下调新生鼠高氧暴露肺组织中IL-6、MCP-1、CINC-1、AR的表达,减轻高氧肺损伤的肺水肿程度。 Objective To evaluate the effects of red sage root on the expression of cytokines in hyperoxia-induced lung injury in neonatal rats. Methods Aged 2 days old newborn SD rats were randomly assigned to 4 groups of air plus normal saline (NS) (group I ), air plus red sage root (group Ⅱ ),hyperoxia plus NS(group Ⅲ), and hyperoxia plus red sage root(group Ⅳ). On the 14th day after birth, the rats were weighed and the body weight and lung weight were recorded. The expression levels of interleukin-6 (IL-6), monocyte chemoattractant protein-1 (MCP-1), chemokineinduced neutrophilic chemoattractant-1 (CINC-1), and amphiregulin (AR)in the lungs were detected by real-time quantitative-PCR. Results The body weight, lung weight, the ratio of body weight/lung weight, the ratio of wet /dry weight of lung were lower in group Ⅲ than those in group I (P〈 0.05) ,which were higher in group Ⅳ than those in group Ⅲ (P〈0.05). The expression levels of IL-6, MCP-1, CINC-1,and AR mRNA were higher in group Ⅲ than those in group Ⅰ (P〈0.05), which were higher in group Ⅳ than those in group Ⅱ but lower than those in group I (P〈0. 05). Conclusion Red sage root could attenuate the pulmonary edema induced by hyperoxia through reducing the expression levels of IL-6, MCP-1, CINC-1, and AR mRNA in the lung tissues of neonatal rats.
