摘要
目的:探讨痛泻要方的抗炎作用及其机制。方法:以过滤空气皮下注射和1%角叉菜胶囊内注射制备大鼠背部气囊炎症模型预防性灌胃用药,用放免法检测炎性渗出液中IL-6和PGE2,用硝酸还原酶法测定渗出液中NO,用化学发光法分析药物血清对多形核白细胞(PMN)活性的影响。用MTT染色法分析肠上皮内淋巴细胞(IELs)的增殖情况。结果:痛泻要方和氢化可的松均能显著降低大鼠气囊炎症渗出液中IL-6水平,降低大鼠气囊炎症渗出液中NO含量。痛泻要方药物血清、10^-4-10^-6mol/L芍药苷能显著降低PMN的活性,且10^-4mol/L芍药苷的作用与氢化可的松相当,痛泻要方药物血清对PMN活性的影响明显强于正常大鼠血清。痛泻要方药物血清和正常大鼠血清皆能明显促进IELs增殖。结论:痛泻要方具有抗炎作用,其作用机制包括降低炎症渗出液中IL-6、NO水平,抑制PMN活性,促进肠道IELs的增殖。
Objective: The present research aims to investigate the anti-inflammatory effect of Tongxieyaofang and its mechamisna.Methods: To produce rat s back air-pocket inflammation by means of filtered air hypodermic injection and 1% carrageenin injection,to determine IL-6 and PGE2 of the inflammatory exudate through radioimmunoassay,and NO of the exudate through nitrate reductase,to analyze influence of the serum containing drugs on PMN s activity with chemiluminescence approach,and on proliferation of intestinal int:aepithelial lymphocytes (IELs) with MTT method. Results: The inflammatory exudate was collected from the air-pocket; but there was not exudate in the same parts of the rats in the normal control group. The high, medium and low dose of Tongxieyaofang and hydrocortisone were able to reduce IL-6 significantly in the rat airbag inflammatory exudate ( P 〈 0.01 ). The medium dose of Tongxieyaofang and hydrocortisone could significantly reduce NO content in the rat airbag inflammatory exudate (P 〈0.05, P 〈0.01 ). Serum contained drug of Tongxieyaofang, 10^4mol/L- 10^6mol/L paeoniflorin were able to reduce PMN's activity ( P 〈 0.01 ) significantly, and so was able to 10^4mol/L hydrocortisone. The effect of rat serum contained drug of Tongxieyaofang on PMNs'activity was stronger than that of serum from normal control rats ( P 〈 0.05 ). Both serum contained drug of Tongxieyaofang and serum from normal control rats were able to promote IEL proliferation( P 〈 0.01 ) significantly. Conclusion: Tongxieyaofang has anti-inflammatory effect, through reducing the level of IL-6 and NO in inflammatory exudate, suppressing PMN's activity, and promoting proliferation of IELs which plays an important role in protecting intestinal immune barrier.
