摘要
目的:用血清药理学方法观察耳聋左慈汤对庆大霉素(Gentamicin,GM)所致小鼠耳蜗毛细胞损伤的作用。方法:完整分离小鼠耳蜗基底膜,平铺于鼠尾胶上,无血清培养液培养24小时后,正常组更换含10%空白血清的培养液,模型组同时加入0.3mMGM和10%空白血清,耳聋左慈汤药物血清组同时加入0.3 mM GM和10%含药血清,继续培养24小时,免疫荧光染色后荧光显微镜下进行全耳蜗毛细胞计数。用全耳蜗毛细胞损失率评价耳聋左慈汤含药血清对GM所致耳蜗毛细胞损伤的作用。结果:正常耳蜗毛细胞排列整齐,无缺失,而0.3mM GM模型组耳蜗毛细胞大量缺失,外毛细胞损失率(41.8±1.9)%高于内毛细胞损失率(16.8±1.8)%;耳聋左慈汤药物血清可降低耳蜗毛细胞的损失率,内、外毛细胞损失率与模型组比较差异均有统计学意义(P<0.05或P<0.01)。结论:耳聋左慈汤含药血清对GM造成的耳蜗毛细胞损伤有拮抗作用。
Objective: To investigate the serum with Erlong Zuoci Decotion(ELZCD) on gentamicin-induced cochlear hair cells damage.Methods: Cochlear organotypic cultures of newborn mice(postnatal day 2-3) were treated with 0.3mM GM in combination with 10% mice serum or 10% serum with ELZCD.Specimens were treated by TRITIC-phalloidin and examined under a fluorescence microscope.The missing rates of the whole cochlear hair cells were used to evaluate the effects of ELZCD drug-containing serum on GM-induced cochlear hair cells injury.Result: The hair cens in normal cultures arranged regularly without loss. The addition of 0.3 mM GM resulted in significant damage. The missing percentage of OHC(41.8 ± 1.9) % was higher than IHC( 16.8 ± 1.8) %. ELZCD drag-containing serum decreased significantly the missing rates of OHCs and IHCs induced by 0.3mM GM (P 〈 0.05, P 〈 0.01 ). Conclusion: ELZCD drug-containing serum protected cochlear hair ceils from GM induced ototoxity.
