摘要
目的:探讨粉防己碱(Tet)对大鼠心脏移植的抗排斥作用。方法:分离出大鼠外周血单个核细胞(PBMC),用刀豆蛋白A(ConA)和混合淋巴细胞培养(MLC)激活单个核细胞增殖,采用四甲基偶氮唑盐(MTT)比色法法检测单纯应用粉防己碱或粉防己碱+环孢素A(CsA)联合应用后对单个核细胞增殖作用的影响;以W istar大鼠为供者,SD大鼠为受者,建立改良的颈部异位心脏移植模型,根据术后用药不同将其分为4组。①对照组:生理盐水2m l ig(灌胃);②Tet组:每日给予Tet 25 mg/kg ig;③CsA组:每日给予CsA 2 mg/kg ig;④Tet+CsA组:每日给予Tet 25mg/kg+CsA 2 mg/kg ig,观察各组移植心存活时间及病理学改变。结果:①粉防己碱能够显著的抑制单个核细胞增殖,并与CsA有协同作用。②CsA联合粉防己碱显著延长移植物的存活时间,与其它3组比较,差异均有统计学意义(P<0.01)。结论:粉防己碱能够显著的抑制单个核细胞的增殖;粉防己碱联合小剂量环孢素A能明显延长移植心存活时间。
Objective:To investigate the effects of tetrandrine on rejection of allografts of rat heart transplantation.Methods:PBMC was separated from blood of rats,and then actived by ConA,and Mixed lymphocyte culture(MLC).MTT colorimetric assays were used to analyse proliferation of PBMC with tetrandrine or/and CsA;Cervical heart transplants were performed in the disparate rat strain combinations of Wistar donor to SD recipient.The transplanted rats were divided into 4 groups according to different administrations: group 1 : receiving saline 2ml ig;group 2: receiving tetrandrine 25 rag/(kgod) ig; group 3 : receiving CsA 2 rag/( kgod ) ig; group 4 : receiving tetrandrine 25 rag/(kgod) and CsA 2 rag/(kgod) ig. Survival of grafts and histological change were observed. Results:Tetrandrine can greatly inhibit the proliferation of PBMC in dose-dependent manner, and synergistic effects of tetrandrine and CsA were found on PBMC. The combination of tetrandrine and CsA can prolong graft survival significantly ( group 4 vs group 1,2,3 ) (P 〈 0.01 ). Conclusion :Tetrandrine can inhibit the proliferation of PBMC ; Tetrandrine has a synergistic effect with subtherapeutic doses of CsA in prolonging rat cardiac allograft survival.
