应用导流杂交基因芯片技术对HPV感染基因分型检测的研究 被引量:14

摘要 目的探讨核酸分子快速导流杂交基因芯片技术(flow-through hybridization and gene chip,Hybri Max)对人乳头状瘤病毒(human papillomavirus,HPV)感染分型检测中的应用。方法对558例临床患者进行液基细胞学检测(LPT),异常者做组织病理学诊断,同时采用Hybri Max技术,对558例患者的宫颈细胞进行HPV DNA检测和分型。结果558例患者中,细胞学异常的370例,并对异常者做病理学诊断,结果为:慢性炎症80例,CINI70例,CINII76例、CINIII82例、宫颈癌62例。②全部患者均进行HPVDNA的检测与分型,其HPVDNA阳性的是:细胞学正常的188例中有40例(21.3%),慢性炎症有48例(60.0%),CINI有52例(74.3%),CINII有68例(89.5%),CINIII有76例(92.7%),宫颈癌有62例(100%)。结论对HPV感染的检查以及通过Hybri Max技术对HPVDNA基因分型检测的研究,对宫颈癌早期发现,判断预后及指导治疗有重要价值。 Objective To study the application on the detecting genotypes of human papillomavirus infection by flow-through hybridization and genechip.Methods 558 patients were detected by LPT, cases with abnormal cytological diagnosis were detected by the pathological histology, and cervical specimens of 558 patients were detected and typed by using HybriMax. Results 0370 cases with abnormal cytological diagnosis were detected by the pathological histology, the results:80 cases in inflammation, 70 cases in CI- NI,76cases in CINII,82 cases in CINIII,62 cases in cervical cancer.QAll of the patients were detected and typed by using Hybri- Max, the HPV positive results is 40 of 188 cases in normal cytological diagnosis(21.3 % ), 48 of 80 cases in chronic inflanunation(60. 0% ), 52 of 70 cases in CINⅠ(74.3 % ), 68 of 76 cases in CINⅡ(89.5 % ), 76 of 82 cases in CINⅢ(92.7 % ), 62 of 62 cases in cervical cancer( 100% ). Conclusion HPV-DNA detection and HPV-DNA genotyping by using HybriMax are of great importance in early diagnosis, prognosis and treatment of cervical cancer.
