紫菜(Porphyra)叶状体rbcL基因分类学分析 被引量:7

Taxonomy of Porphyra(Rhodophyta,Bangiales) based on rbcL Sequences from the thallus

摘要 对采自江苏海安的坛紫菜(Porphyra haitanensis),浙江南麂的长紫菜(P.dentata)、圆紫菜(P.suborbiculata)和广东汕头的皱紫菜(P.crispata)4种紫菜叶状体rbcL基因进行扩增及序列测定,并用NJ法构建系统树.序列分析结果表明,4种紫菜rbcL扩增序列长度均约为1400bp,其中皱紫菜与坛紫菜仅6个碱基差异位点.距离矩阵结果显示,各紫菜种内遗传距离在0.000~0.010之间,遗传距离最小的是坛紫菜,遗传距离最大的是圆紫菜.各紫菜种间的遗传距离在0.001~0.094之间,其中遗传距离最小的是皱紫菜与坛紫菜,遗传距离最大的是圆紫菜与皱紫菜、条斑紫菜与圆紫菜.系统树的聚类分析结果显示,坛紫菜与皱紫菜亲缘关系最近,与长紫菜形成姊妹分支,条斑紫菜和圆紫菜依次与之相聚.本研究利用紫菜rbcL序列差异可区分紫菜种间关系,结果表明系统发育地位在紫菜种间关系中有更重要的影响,结果并不支持根据边缘进行的分类系统. The rbcL genes of Porphyra haitanensis from Haian, Jiangsu Province, P. dentata and P. suborbiculata from Nanji, Zhejiang Province and P. crispata from Shantou, Guangdong Province were amplified and sequenced in the present study. The analysis results of sequences showed the length of amplified fragments from four species were about 1400bp. The intraspecies and interspecies pairwise distance ranged from 0.000 to 0.010 and 0.001 to 0.094, respectively. P. haitanensis and P. crispata were clustered together in phylogenic tree based on rbcL genes forming a sister group of P. dentata. Then P. yezoensis and P. suborbiculata was clustered in turn. The results of present study indicated that rbcL gene can be useful in elucidating the relationship between different species of the genus Porphyra. The phylogenic position of certain species of Pophyra was a more important factor than geological separation that affects the relationship between different species. The results did not support the taxonomy system based on the type of marginal parts of Porphyra thallus.
