糖络宁对STZ诱导糖尿病大鼠血清IGF-1含量及肝组织IGF-1基因表达的影响 被引量:4

Influences of Tangluoning Oral Solution on content of serum IGF-1 and gene expression of liver IGF-1 in rats with diabetes induced by STZ

摘要 目的观察中药糖络宁对糖尿病大鼠血清胰岛素样生长因子-1(IGF-1)含量及肝组织IGF-1基因表达的影响。方法采用链脲佐菌素(STZ)诱导糖尿病大鼠模型,造模成功后随机分为模型组、西药组(依帕司他)、中药糖络宁组,共给药12周,给药结束后,用ELISA法测定大鼠血清IGF-1含量,用RT-PCR法测定大鼠肝组织IGF-1基因表达。结果与正常组大鼠比较,模型组大鼠血清IGF-1含量明显下降、肝组织中IGF-1 mRNA表达明显下调(P<0.01);与模型组大鼠比较,西药组和糖络宁组大鼠血清中IGF-1含量明显增高(P<0.01或P<0.05),糖络宁组与西药组比较也有显著差异(P<0.01);与模型组大鼠比较,糖络宁组和西药组可提高大鼠肝组织中IGF-1 mRNA的表达(P<0.01)。结论糖络宁可提高糖尿病大鼠血清IGF-1水平,上调糖尿病大鼠肝脏IGF-1 mRNA表达。这可能是糖络宁治疗糖尿病周围神经病变的机制。 Objective To observe the influences of Tangluoning Oral Solutlon (TOS) on me content of serum insulin-like growth factor-1 ( IGF-1 ) and gene expression of liver IGF-1 in rats with diabetes induced by streptozotocin (STZ). Methods The rat model of diabetes was established by the induction of STZ, and then all rats were randomly divided into the model group, Western drug (epalrestat) group and TOS group. After treatment for 12 weeks ELISA was used to detect the content of serum IGF-1 and RT-PCR was applied to determine the expression of liver IGF-1. Results In the model group the serum 1GF-1 content decreased significantly and the expression of liver IGF-1 mRNA was down-regulated significantly compared with those in the normal group ( P 〈 0. 01 ). In Western drug group and TOS group serum IGF-1 content increased significantly compared with that in the model group respectively (P 〈0.01 or P 〈0.05). There was a significant difference between TOS group and Western drug group (P 〈 0.01 ). In TOS group and Western drug group the expression of liver IGF-1 mRNA increased (P 〈0.01 ) compared with that in the model group. Conclusion TOS can increase serum IGF-1 level, up-regulate the expression of liver IGF-1 mRNA, which may be the mechanism of TOS for treating diabetic peripheral neuropathy.
