RP-HPLC同时测定银翘解毒片中绿原酸、连翘苷和甘草酸的含量 被引量:12

Quantitative determination of chlorogenic acid,forsythin and glycyrrhizic acid in Yinqiaojiedu Tablets by RP-HPLC simultaneously

摘要 目的研究银翘解毒片中绿原酸、连翘苷和甘草酸的含量和测定方法。方法采用反相高效液相色谱法,色谱柱为Diamonsil^TM C18(250mm×4.6mm,5μm),乙腈-0.1%磷酸水溶液为流动相,梯度洗脱,流速为1.0mL/min,三波长检测,λ326nm,λ2为228nm,λ3为252nm,柱温30℃。结果绿原酸、连翘苷和甘草酸进样量分别在0.3956~2.7692μg、0.5928—4.1496μg和0.5583—3.9080μg范围内呈良好的线性关系,相关系数分别为0.9991、0.9998和0.9999,平均加样回收率分别为100.5%、97.1%和99.6%,RSD分别为1.6%、2.1%和1.2%(n=6)。结论该方法专属性好、准确度高,可用于综合评价银翘解毒片的质量。 Objective To study the contents and determination method of chlorogenic acid, forsythin and glycyrrhizic acid in Yinqiaojiedu Tablets. Methods The procedure of RP-HPLC was performed on a column of Diamonsil^TM C18 (250 mm ×4.6 mm, 5 μm) using acetonitrile-0.1% phosphoric acid water solution as the mobile phase in gradient elution. The flow rate was 1.0 mL/min,λ1 was 326 nm, λ2 was 228 nm, and λ3 was 252 nm in three-wavelength detection. The column temperature was 30℃. Results The sample size of chlorogenic acid, forsythin and glycyrrhizic acid showed respectively a good linear relationship at range of 0.395 6 -2.769 2 μg, 0.592 8 -4. 149 6 μg and 0.558 3 -3.908 0 μg (r= 0.999 1, r=0.999 8 and r=0.999 9). The average recovery (n=6) was 100.5%, 97. 1% and 99.6% (RSD = 1.6%, 2. 1% and 1.2% ) respectively. Conclusion The method is peculiar with a good reproducibility, which can be used for the qualitative evaluation of Yinqiaojiedu Tablets.
