低温冻存对人牙周膜干细胞生物学特性的影响 被引量:6

摘要 目的观察超低温冻存对人牙周膜干细胞(PDLSCs)生物学特性的影响。方法体外分离、培养人牙周膜细胞,待细胞达一定量后用有限元稀释法筛选PDLSCs。将第3代人PDLSCs经液氮冻存半年后复苏,测其存活率,检测细胞免疫表型,对PDLSCs进行成骨诱导,测定其碱性磷酸酶(ALP)活性及体外矿化能力,观察超低温冻存对PDLSCs生物学特性的影响。结果第3代人PDLSCs冻存、复苏前后两组细胞在细胞形态、生长过程、免疫表型、成骨分化能力等方面均无显著性差异(P>0.05)。结论超低温冻存对人PDLSCs活性、增殖、细胞免疫表型及成骨分化能力没有明显影响。 Objective To study the effects of cryopreservation on biological characters of human periodontal ligament stem cells (PDLSCs). Methods Periodontal ligament cells was obtained from healthy young human teeth extracted for orthodontic reason. PDLSCs were isolated by limiteA dilute method. PDLSCs were cryopreserved in liquid nitrogen for 6 months. The viability, proliferative abilities, immunophenotype, osteogenic differentiation ability of thawed PDLSCs were investigated and compared with these of precryopreserved PDLSCs. Results There were no differences between two PDLSCs groups in the cell survival rate, proliferative abilities, immunophenotype, osteogenic differentiation ability. Conclusion The biological characteristics of PDLSCs have no changes alter cryopreservation.
