摘要
目的研究FAT10 mRNA在结直肠癌组织中的表达及其与结直肠癌临床病理学间的关系。方法应用逆转录聚合酶链反应法(RT-PCR)检测FAT10 mRNA在51例结直肠癌组织、结直肠癌旁组织和正常结直肠黏膜组织的表达水平,并观察其与临床病理特征的关系。结果(1)RT-PCR法检测FAT10在结直肠癌组织病例中表达阳性率为68.6%,在结直肠癌旁组织及正常结直肠黏膜中表达阳性率分别为17.6%和13.7%;(2)结直肠癌组织FAT10表达水平与结直肠癌旁组织及正常结直肠黏膜组织比较,差异均有显著性(P<0.05);结直肠癌淋巴结转移组患者FAT10表达水平与未转移组相比较,差异有显著性(P<0.05);伴肝转移组的结直肠癌与无肝转移组结直肠癌FAT10表达水平差异有显著性(P<0.05);组织FAT10表达水平还与临床分期密切相关,FAT10水平随分期升高而升高。与患者年龄、性别、肿瘤所在部位、肿瘤大小及病理分化程度等,临床病理特性之间无相关性(P>0.05)。结论FAT10在结直肠癌组织中的表达明显上调,对应的癌旁组织及正常结直肠黏膜组织无表达或弱表达;测定FAT10的表达可能预测结直肠癌的发生潜能;结直肠癌组织中FAT10的阳性表达与肿瘤临床分期、有无淋巴结转移及肝转移有关,提示FAT10基因与肿瘤的侵袭、转移密切相关。
Objective To study the expression of FAT10 in colorectal cancer tissue. And analysed the relationship between the expression of FAT10 and the character of clinical pathology in colorectal carcinomas. Methods FAT10 mRNA expression in 51cases of colorectal carcinoma , paracancerous tissue and their normal colorectal tissue were detected by using reverse transcription- polymerase chain reaction(RT-PCR). The relation between FAT10 mRNA expression and the character of clinical pathology were observed. Results ( 1 ) By RT - PCR method, the positive expression rates of FAT10 in colorectal carcinomas were 68.6%, the positive expression rates of FAT10 in paracancerous tissue and normal colorectal tissu were 17.6% and 13.7%. (2)The levels of FAT10 in tissue of colorectal carcinomaswere obviously higher than that of paracancerous tissue group and normal colorectal group( P 〈 0.05). Compared with the group of no lymph node metastasis, the FAT10 levels of lymph node metastasis group was obviously higher( P 〈 0.05) .The intensity of FAT10 expression in tissue of colorectal carcinomas was correlated with liver metastasis( P 〈 0.05). The FAT10 levels were positively related with clinical stages. And the levels of FAT10 had ascending trends according to the clinical stages. The FAT10 levels were no correlated with age, sex, the position of tumor occurrence diameter of tumor, the extent of polarization and other clinicpathological factors. Conclusion Expression of FAT10 was highly upregulated in colorectal cancer, but not in the paracancerous tissue group and normal colorectal group. The abnormal expression of FAT10 may play important roles in the tumorigenesis and progression of colorectal carcinoma. Expression of FAT10 had the association with lymph nodes metastasis, liver metastasis and the clinic type of pathology. It may have the important significance on forecasting the invasion and metastasis of colon carcinomas.
