摘要
目的观察慢性束缚应激大鼠海马及杏仁核谷氨酸受体1(GluR1)、谷氨酸受体2(GluR2)表达的变化及逍遥散对其的影响。方法雄性SD大鼠30只,按体重随机分为3组,每组10只:A组,正常对照组;B组,模型组;C组,逍遥散组。B、C组以慢性束缚方法制作应激模型,A组散养于各自饲养箱。自造模第1天开始,C组每日在束缚前1 h灌服逍遥散有效组分悬液;A、B组灌服等量蒸馏水。造模21 d结束后,各组大鼠麻醉后灌注取脑,冰冻切片后行GluR1、GluR2免疫组织化学染色。结果模型组的GluR1在海马CA1和DG区表达增多;GluR2在CA1、CA3和DG区表达下降;GluR1和GluR2在基底外侧杏仁核(BLA)区均无明显变化。逍遥散可下调GluR1阳性细胞及免疫阳性物表达,上调GluR2阳性细胞数及免疫阳性物表达。结论肝郁脾虚证模型大鼠α-氨基羟甲基恶唑丙酸(AMPA)受体亚基GluR1和GluR2在海马的表达趋势不同;海马CA1区可能是肝郁脾虚证模型大鼠突触可塑性的最敏感部位;逍遥散对GluR1和GluR2表达的调节呈现出区域选择性、时相性和双向调节作用。
Objective To observe the changes of glutamate receptors 1 ( GluR1 ) and glutamate receptors 2 (GluR2) expressions in hippocampus and amygdala in rats of chronic restraint stress, and the influence of Xiaoyao Powder. Method Male SD rats ( n = 30) were randomly divided into three groups ( each n = 10), including the normal control group (group A), model group (group B) and Xiaoyao Powder group (group C). Group B and C were given chronic restraint to establish the stress model, and group A was scatter-fed in breeding box. From the first day of modeling, group C was fed daily the suspension of effective components of Xiaoyao Powder one hour before the stress, andgroup A and B were fed equivalent distilled water. After 21 days rats in all groups were anaesthetized and affused. The rat brain was collected, frozen, sliced up, and then stained by immunohistochemistry method. Result In the model group the expression of GIuR1 was increased in regions of CA1 and DG, and expression of GluR2 was decreased in regions of CA1, CA3 and DG. The expressions of GluR1 and GluR2 did not change in BLA region. Xiaoyao Powder could down-regulate the expressions of GluR1 positive cells and immune positive substance, and up-regulate the expressions of GluR2 positive cells and immune positive substance. Conclusion The expressions of GluR1 and GluR2 of amino-3-hydroxy-5-methyl-4-isoxazole propionic acid (AMPA) in hippocampus had different trends in the rat model of liver-stagnation and spleen-deficiency syndrome. CA1 region of hippocampus may be the most sensitive region of synaptic plasticity. The regulation effects of Xiaoyao Powder on the expressions of GluR1 and GluR2 show the characteristics of regional-selective, time-phase and double-directional.
