摘要 目的分离、培养和纯化大鼠切牙颈环上皮细胞,观察牙釉蛋白在大鼠切牙颈环上皮细胞中的表达情况。方法取出生2d的威斯塔大鼠牙胚颈环组织,培养原代混合细胞,纯化颈环上皮细胞,将细胞进行抗牙釉蛋白免疫细胞化学染色。结果牙釉蛋白在培养且纯化的颈环上皮细胞中呈强阳性表达,对照组呈阴性表达。结论大鼠切牙颈环上皮细胞具有向成釉细胞分化的潜能。 Objective To isolate and culture and purify epidermal cells from rat cervical-loop, and to observe the expression of amelogenin in epidermal cells. Methods Tissues obtained from lower incisor of postnatal 2-day Wistar rats'cervical-loop were cultured in the Dulbecco minimum essential medium(DMEM)-F,2 medium. The cervical-loop epidermal cells grew confluent after purified from fibroblast-like cells by differential digestion. Then the purified cells were immunocytochemical stained with antiamelogenin antibody. Results The primary culture was exhibited positive staining to amelogenin while the control group was negative. Conclusion The epidermal cells isolated from rat cervical-loop epithelium have the potentiality to differentiate into ameloblasts.
