水飞蓟素对肾小管间质纤维化大鼠CTGF的影响 被引量:2

摘要 目的探讨水飞蓟素对肾间质纤维化大鼠结缔组织生长因子(CTGF)表达的影响。方法56只雄性SD大鼠,随机分为假手术组,梗阻组,水飞蓟素治疗组。以单侧输尿管结扎术建立输尿管梗阻的动物模型。术后第7、14、21天取术侧肾组织观察病理变化并应用免疫组织化学方法检测肾组织CTGF的表达。结果水飞蓟素可以显著改善梗阻侧肾组织病理变化,下调CTGF表达。结论水飞蓟素可能是通过下调CTGF表达而抑制纤维化的发生,从而起到对肾脏的保护作用。 Objective To investigate the effects of silymarin on the expression of connective tissue growth factor (CTGF) in the kidneys of rats with unilateral ureteral obstruction (UUO)-induced renal fibrosis. Methods Fifty-six rats were randomly divided to shame operation group, operation group and silymarin [ 30mg/( kg· d) ] treatment group after unilateral ureteral obstruction. Tubulointerstitial fibrosis model was estab- lished via unilateral ureteral obstruction (UUO). Renal tissues were examined by light microscopy 7 th, 14 th,21 th days after operation. Immunhistochemistry was applied to measured the expression of CTGF in the renal interstitium. Results Silymarin may significant ameliorate pathological change of nephridial tissue after unilateral ureteral obstruction. Silymarin may down regulate the expression of CTGF in renal tissue. Conclusion Silymarin may inhibit renal fibrosis by down-regulating CTGF expression, which may play a key role in protection against renal fibrosis.
