摘要
目的探讨C反应蛋白(CRP)对多发性骨髓瘤细胞株U266细胞株生长的影响及其机制。方法将CRP(浓度为0、5、10、20mg/L)与U266细胞株作用24h后,血液分析仪检测细胞增生情况;采用反转录-聚合酶链反应(RT—PCR)方法检测凋亡相关基因Survivin和HSP-90α的表达。结果培养24h后,U266细胞增生率明显提高(P〈0.05),且与CRP浓度成正比,CRP浓度为20mg/L时,细胞增生最明显;U266细胞表达Survivin、HSP-90α,其表达量随CRP浓度的增高而增高(P〈0.05)。结论CRP可促进U266细胞株增生,并通过调节凋亡抑制蛋白Survivin及HSP-90α表达的增高发挥其诱导U266细胞增生的作用。
Objective In the present study, we examined the expression changes of Survivin/HSP90α, in C-reactive protein(CRP) treated human muhiple meyloma (MM) cell lies. Methods The human MM cell lines were cultured for 3 days and incubated with human CRP(0.5, 10, 20 mg/L for 24 hours) and analyzed by blood analyser for proliferation ratio. The effects of CRP in MM cell lines in various concentrations on survivin/HSP-90α mRNA expression were examined by RT-PCR. Results Proliferation ratio of U266 cell line was signitieantly increased, and the expression of survivin/HSP-90α were upregulated (gene promoting cell survival) at mRNA levels in proportion to increased CRP concentrations. Conclusion These results demonstrated that proliferation ratio, survivin and HSP-90α could be regulated by CRP in MM cells.
