放射线对口腔鳞状细胞癌细胞中核因子-κB p65表达的影响

Effected of radioactive ray on the expression of NF-κB p65 in human oral squamous cell carcinoma

摘要 目的:探讨不同剂量放射线对口腔鳞状细胞癌(OSCC)细胞中核因子-κB(nuclear factor-κB,NF-κB)p65信号转导蛋白表达的影响。方法:采用人舌鳞状细胞癌Tca8113细胞,分为6个剂量组,分别为0y、2、4、6、8、10Gy,在照射后的不同时间点(1,3,6,10,24,48h)应用免疫细胞化学检测NF-κB p65的表达情况,应用Leica Qwin Plus图像分析系统对图像进行分析,测其灰度值,并应用SPSS10.0软件对数据进行X2检验。结果:免疫细胞化学方法显示不同剂量组的平均灰度值与0Gy组相比均为P<0.05,差异均具有统计学意义,而不同时间点组的平均灰度值相互比较,3h组的平均灰度值与其他时间点组相比均为P<0.05,差异有显著性。结论:放射线可以激活NF-κB p65信号转导通路,此信号转导通路在肿瘤的放射治疗中可能具有重要意义。 Objective:To investigate the effected of radioactive ray on the expression of NF-κB p65 in human oral squamous cell carcinoma. Method:To adopt the squamous cell carcinoma of tongue Tca8113 cell,the experiment was divided into six groups,0、2、4、6、8 and 10 Gy group. To use immunocyteochemistry to detect the NF-κB p65 protein after irradiation in various times(1、3、6、10、24、48 h) .To apply the Leica Qwin Plus image analytical system to analyzed the gray scale. Result:To compare the brtween various dose group and 0 Gy group ,the P 〈0.05,the difference was statistical significance.The difference of average gray scale between 3h group and other different time group was significance (P 〈0.05). Conclusion: Radioactive ray can activitate the NF-κB p65 of oral squmaous cell carcinoma cell.NF-κB p65 maybe an important activity in the tumor radiotherapy.
