奥扎格雷钠对缺血再灌注大鼠脑海马区细胞保护作用的研究 被引量:9

摘要 目的观察奥扎格雷钠对大鼠脑缺血/再灌注后IL-6和TNF-α蛋白表达的影响,探讨奥扎格雷钠对缺血再灌注大鼠脑海马区细胞的保护作用及其作用机制。方法将40只SD雄性大鼠随机分为假手术组、缺血/再灌注组、生理盐水对照组和奥扎格雷钠治疗组。采用大鼠大脑中动脉线栓(MCAO)法制备局灶性脑缺血/再灌注模型,应用免疫组化技术分别检测各组大鼠脑组织IL-6和TNF-α蛋白表达情况。结果与假手术组相比较,脑缺血/再灌注组IL-6和TNF-α蛋白的表达明显增多(P<0.05),奥扎格雷钠能够明显减少TNF-α的表达(P<0.05),而对IL-6表达无影响(P>0.05)。结论奥扎格雷钠对大鼠脑缺血/再灌注具有保护作用,其机制可能与抑制TNF-α表达有关。 Objective To observe the effect of ozagrel sodium on the levels of IL-2 and TNF-α in rats with cerebral ischemia-reper- fusion injury, and study the mechanisms of protective effect s of Ozagrel sodium against focal cerebral ischemia-reperfusion injury in rats. Methods The rats were randomly divided into sham, ischcmia-reperfusion, saline and ozagrel sodium groups. The right middle cerebral artery of the rats were occluded by inserting a thread through internal carotid artery for 2 h, and then reperfused for 12 h. Immunohistochemical techniques were used to detect IL-6 and TNF-α protein expression in the brain tissue of rats. Results The levels of IL-6 and TNF-α were significantly increased in the brain tissue of isehemia-reperfusion group compared with those of sham group. Compared with that of ischemiareperfusion group, ozagrel sodium significantly reduced the level of TNF-α( P 〈 0. 05 ) , and had no effect on IL-6 expression (P 〉 0.05 ). Conclusions Ozagrel sodium can reduce ischemia-reperfusion injury by decreasing the damage to the TNF-α.
